Medical and Diagnostic Applications
Infection Diagnosis: RT-PCR is a primary tool for detecting viral infections, such as COVID-19, influenza, HIV, and Hepatitis C, by identifying the viral RNA in patient samples. Cancer Monitoring: It helps track cancer progression and treatment response by measuring changes in gene expression levels in tumors. Genetic Disorder Screening: The technique can detect specific genetic mutations associated with certain inherited conditions, such as Lesch-Nyhan syndrome.

Biological Research Applications
Gene Expression Profiling: RT-PCR allows for the precise quantification of gene expression levels to study gene function and cellular processes. RNA Interference (RNAi) Validation: Researchers use it to confirm that target genes have been effectively "knocked down" in RNAi experiments. Cloning and DNA Manipulation: The process generates complementary DNA (cDNA) from RNA, which can then be cloned and used in further molecular biology studies.

Food Safety and Environmental Monitoring
Pathogen Detection: In food and environmental testing, RT-PCR rapidly detects bacterial and viral pathogens like Salmonella, Listeria, and E. coli in food, water, and soil samples. GMO Detection: It is used to identify the presence of genetically modified organisms (GMOs) in food products. Ingredient Authentication: RT-PCR can also confirm the presence or absence of specific ingredients, such as pork in halal or kosher foods.

  1. Excitation:
A light source, often ultraviolet light, is directed at the sample.
  2. Excitation State:
Molecules in the sample absorb photons of light and enter a higher energy "excited state".
  3. Emission:
As the excited molecules return to their ground state, they release the absorbed energy by emitting a photon of light. This emitted light is the fluorescence.
  4. Detection:
A fluorescence detector (a type of spectrofluorometer) measures the intensity and wavelengths of the emitted light.
  5. Spectra:
The data collected can be plotted as:
    • Fluorescence Emission Spectrum: Shows the intensity of emitted light at different wavelengths for a fixed excitation wavelength.
    • Fluorescence Excitation Spectrum: Shows fluorescence intensity at a specific emission wavelength across a range of excitation wavelengths.
    • Excitation-Emission Matrix (EEM): A 2D plot of fluorescence intensity as a function of both excitation and emission wavelengths.

Fast Protein Liquid Chromatography, a laboratory technique used to separate and purify proteins, peptides, and other biomolecules. It utilizes a stationary phase of resin-filled columns and a mobile liquid buffer to achieve high-resolution separation based on various properties, such as size, charge, or affinity. FPLC operates at medium pressure and employs automated systems for buffer control, sample injection, and fraction collection, enabling efficient and reproducible purification of biomolecules.

Biomolecule Purification
  • Proteins & Peptides: FPLC is widely used to purify specific proteins and peptides from complex mixtures.
  • Nucleic Acids: It can also be used for the rapid purification of RNA, synthetic oligonucleotides, and plasmid DNA.
    • Clinical Applications
      • Disease Diagnosis: FPLC can separate proteins in bodily fluids like cerebrospinal fluid (CSF) and urine to identify disease markers.
      • Protein Profiling: It is used to analyze protein profiles for conditions such as tubular proteinuria and to detect changes in CSF protein composition.
      • Diagnostic Markers: FPLC helps in the diagnosis of diseases like beta-thalassemia by separating specific blood proteins, according to Unacademy.
      Research & Biotechnology
      • Drug Discovery: The technique is employed in drug development processes to separate and analyze biomolecules.
      • Enzymology: FPLC is a valuable tool for isolating, purifying, and characterizing enzymes.
      • Recombinant Protein Production: It is used to purify recombinant proteins and antibodies for various uses.
      Other Applications
      • Food Industry: FPLC is used to analyze the nitrogenous constituents in beer, which are relevant to its foam stability.
      • Pest Control: The technique has been used to purify animal venoms for various applications.
